Abstract

In this research a streamlined and economical UV spectrophotometric approach have been successfully developed to simultaneously measure triprolidine hydrochloride (TRI) and pseudoephedrine hydrochloride (PSE). This analytical strategy was specifically designed to handle both bulk raw materials and pharmaceutical tablet formulations, offering a practical solution for laboratories that require high-quality results but may have limited access to complex instrumentation like HPLC. The study introduced two distinct mathematical methods to resolve the common challenge of overlapping light absorption spectra between these two drugs. Both techniques utilize 0.1 M hydrochloric acid as the solvent and rely on absorbance readings taken at specific wavelengths of 257 nm and 290 nm. Method A establishes a relationship based on concentration and absorbance ratios, while Method B applies an absorbance factor to mathematically correct for the spectral overlap of the components. To ensure the reliability of these procedures, both methods were rigorously validated according to ICH guidelines. The results demonstrated a high degree of linearity, with correlation coefficients exceeding 0.999 for both TRI (at concentrations of 6–30 μg/mL) and PSE (at 144–720 μg/mL). Furthermore, the methods proved to be highly precise, maintaining a relative standard deviation of less than 2%, and accurate, with recovery rates consistently near 100%. Statistical analysis confirmed that there is no significant performance difference between the two proposed techniques. Consequently, these UV spectrophotometric procedures serve as a simple, dependable, and efficient tool for the routine quality control of triprolidine and pseudoephedrine combinations in pharmaceutical settings.
